Cam Timing

Is it possible to be off by one tooth on cam timing and the bike still run well but have lackluster performance in the high rpm range ( 7k and up) ? I upgraded to an actual 250cc cyl, thin head gasket and ported the head. low and mid rpm performance is great but high rpm feels maybe a hair better than stock MAYBE. I am working on getting a tune and I did put everything in time but I thought maybe the marks were off a little , just looking at theories.
