Yes, you must set the valve lash at TDC on the compression stroke.

Here is a foolproof method for finding TDC on the compression stroke.

Remove the two inspection ports at the top of the engine, and rotate the crankshaft in the counter-clockwise direction. Look for the "L" mark on the cam sprocket. After you see the "L" mark, carefully rotate the crankshaft until the "L" mark is aligned with the left inspection port, and the "R" mark is aligned with the right inspection port. When the cam sprocket is in this position, you are always aligned with TDC on the compression stroke.

You don't even need to open the inspection port on the flywheel. Using this method, you will never make a mistake by adjusting the valves on the exhaust stroke, instead of the compression stroke.
